2
Q

what are the limitations of the MM plot

A

using MM graph makes the determination of vmax and km inaccurate due to hyperbolic plot

vmax is approached but not attained

Q

what is the effect of a competitive inhibitor on the michaelis menten plot and LB plot

A

vmax is unaffected, km is increased.

no effect on the vmax value bc the addition of more substrate can overcome the effect of the comp inhibitor

km increases as there is increased competition between the substrate and the comp inhibitor

Q

what is the effect of a noncompetitive inhibitor on an LB plot and MM plot

A

vmax is lowered, km is unaffected

vmax is lowered bc the enzyme substrate inhibitor complex is unable to form any product

km is unaffected bc the enzyme substrate complex will form bc the inhibitor will bind to the allosteric site

Q

What is the effect of pH on enzymes?

A

enzyme tertiary structure held together in part by ionic bonds

at high/low pH ionic bonds become disrupted = change structure
