1

To do linkage analysis, what are the 2 things you need to do?

genotype lots of family memebers
Haplotype pairs of alleles inherited together

2

What are used to detect polymorphisms?

RFLP, VNTR, Microsatellites, SNP (most used)

3

What kinds of tests are these?
- DNA microarrays,
- allele specific oligonucleotides (ASO),
- single-base extension sequencing,

SNP tests

4

What do you have to do with RFLP and VNTR tests?

utilized southern blots and PCR

5

Microsatellite tests use what?

PCR

6

Is hunington and Neurofibrosis 1 dominant or recessive?

dominant

7

Is PKU and Neurofibrosis 1 dominant or recessive?

recessive

8

How can you determine whether recombination has occured?
When you have recombin between two alleles that are close, will the events be rare?

use several polymorphic markers that closely flank the gene
yes, because they would still be inherited together if they were really close i.e. you will still have the marker.
