Microscopy and Histometry Flashcards
(8 cards)
Describe the pathway light travels from light source to the eye for polarising microscopes
light soucre-> condenser lense-> polariser-> specimen-> analyser-> objective lens-> eyepiece
Which special feature does light pass through in a dark field microscope?
Dark field condenser with a central opaque disc
Which special feature does light pass through in a polarising microscope?
Polariser- below specimen stage
Which special feature does light pass through in a fluorescent microscope?
Excitation filter followed by a dichroic mirror then finally an emission filter
Which special feature does light pass through in a phase contrast microscope?
phase platea and a condenser annulus
Which special feature does light pass through in a interferance microscope?
beam splitter, interferacne rings/ optical compensators
Which common optical illusions can affect our judgement when validating tissue sections
- Chromatic aberration- colour fringing cn blur tissue structures
- Out of focus blur- tisues can appear distorted
- Colour perception variability- differences in colour perception due to lighting
How is the stage micrometre and eyepiece micrometre (graticule) combination used to measure structures in tissue sections?
allows for measurement of objects at the microscopic level by using the collaborative scale in the eyepiece in conjunction with a known reference scale on the microscope
