Model the 2 neuron circuit. First neuron is Gaba and the second is DA. What are the important receptors on them? (hint: three on both)
Gaba: Gaba A, CB1 terminal, and MOR
DA: Gaba A, nACh, and DAT receptors
What are the equilibrium potentials for all four important ions of the neuron?
K+: -88mV
Na+: +60 mV
Cl-: -61 mV
Ca+: 134 mV
Run through all the phases of an action potential
Resting: cell is at -70 mV
Depolarization: cell decreases in voltage to -55 mV, causing the VG-Na+ to influx into the cell.
Repolarization: once +40mV is reached, K+ channels are allowed to open and K+ outflux ensues
Refractory period and hyperpolarization: as the voltage decreases, the Na+ stay closed until -55mV is reached, where they then open during hyperpolarization
What is the differences between Gi, Gs, Gq, and G12/13 receptors both in stucture and action. What important receptors belong to each and how does the receptor work?
-The receptor complexes only differ in unit subtype
-Gi: inhibitory. These increase K+ outflux and decrease Ca++ influx. Includes MOR, CB1, D2 thru D4, and some 5HT
-Gs: stimulatory, increases Na+/Ca++ influx
Includes D1, D5, TARR (amphetamines), and some 5HT
-Gq: stimulatory, releases Ca++ from stores
Includes 5HT2A
*G12/13: start cytoskeletal rearrangment
What are the drugs that target GABA-A receptor and what is their MOA?
- EtOH: possitive allosteric modulator at GABA-A terminols on GABA-A cells
- Sedative: PAM at GABA-A receptor (same as EtOH)
What drugs target the MOR receptor and what is their MOA?
MOR is a Gi receptor that is targeted by opioids. Opioids work as an agonist at the receptor, downregulating GABA neurons.
What are the drugs that target CB1 receptor and what is their MOA?
Cannabinoids are agonists at the CB1 receptor on the GABA neuron, which is a Gi receptor
What subunits of the GABA-A channel does have to have for EtOH to work? What subunit can it not have for Zolpidem to work?
EtOH needs the delta receptor.
Zolpidem cannot have the gamma receptor.
What are the drugs that target nACh receptor and what is their MOA?
Nicotine is an agonist at the stimulatory ligand gated channel nACh on the DA neruons
What drugs target the DAT on dopamine neurons and what is its MOA?
cocaine and meth. They inhibit the reuptake of DA from the DA neuron by blocking the DAT.
