Q

Other methods for platelet count

A

1.Reese-Ecker Method
2.Ammonium Oxalate Technique
3. Direct Method
- Nygard’s
- Guy and Leake’s
- Brenker-Cronkite
- Unopette
4. Indirect Method
- Dameshek
- Fonio’s
- Olef’s

17
Q

Diluting fluid composition of reese-ecker method

A

•brilliant cresyl blue
•40% formalin
•sodium citrate

18
Q

Procedure of reese-ecker method

A
  1. Draw blood up to 0.5 mark of the rbc pipette
  2. Add reese-ecker diluting fluid up to 101 marks
  3. Shape the pipette for 1 to 5 minutes
  4. Discard 5 to 6 drops and discharge the counting chamber
  5. Place the counting chamber on a petri dish with a wet filter paper to prevent evaporation. Let it stand for 10 to 15 minutes to allow the platelet to settle
  6. Count the plate that’s in all 25 tertiary squares of central secondary square
  7. Compute the platelet count
19
Q

Computation for reese ecker method

A

Number of cells / mm3 = total number of cells counted/areaxdepthxdilution

Or

Cells counted x 2000

20
Q

Procedure of ammonium oxalate technique

A
  1. Draw blood up to the 1.0 mark of the rbc pipette and ammonia oxalate up to the 101 marks
  2. Procedure 2 to six of reese ecker method perform
21
Q

Computation for ammonium oxalate technique

A

Number of cells/mm3 =total number of cells counted/area x depth x dilution

Or

Cells counted x 1000

22
Q

Direct methods of platelet count

A

•Nygard’s
•Guy and Leake’s
•Brenker-Conkrite

(NGB)

23
Q

Indirect method of platelet count

A

•Dameshek
•Fonio’s
•Olef’s

(DFO)

24
Q

The best indirect method of platelet count

A

Olef’s Method

25
Sources of error of platelet count
•platelet clumps •imperfect sources of blood •blood in edta kept at 20°C
