Q

What is the main idea used by genome sequencing tech?

A

Short sequences MANY times, overlap and align based on matching sections assemble into larger sequences then into Chromosome or genomes.

Q

What are Contigs and Scaffolds?

A

Contigs are long assembled sequences, while Scaffolds are contiguous, ordered contigs.

Scaffolds can then be added together to create chromosomes or entire genomes

Q

What is the differences between Shotgun and ordered assembly?

A

Shotgun = breaking up a DNA molecule randomly and then finding overlapping fragments to then build up a sequence
Ordered assembly = a set of overlapping cloned fragments

Q

Pyrosequencing

A

generates 150 bp per reaction
No electrophoresis or other type of fragment separation is needed
Faster than chain termination
