A
open lid of petri dish as little as possible- prevent contamination of apparatus with unwanted microorganisms
wear gloves and mask- prevent cont from bacteria on hands or in the air from breath
flame the loop- maintain pure culture of bacteria
boil agar- so no contamination other than bacteria

random mutation occurs (not due to antibiotic) produces resistant allele/gene resistant bacteria survive and reproduce increases frequency of resistance allele example of directional frequency
