what is complementation?
A recessive mutation in different genes mans that they complement each other and make the WT
- when mutations do not complement each other they are on the same gene
how can you interpret complementation tables?
Identifying complementation groups
- no complementation
+ complementation
Looking horizontally to see where they match up (- at the same spot = same group) (- at different spots in different groups)
in the bacteriophage T4 experiment what did k (lambda) and B strain tell you ?
-WT (not in the same group) can infect B strain and K (lambda)
-mutation can only affect B strain1
How can you get complementation with only one copy?
Dual infection: simultantiously infect bacteria with 2 different bacteriophages and both chromosomes are injected into E. coli and recombination can occur (the b strain infects the rll- and rll+)
how can you check to see if mutations are on the same gene?
check the mutation by complementation testing
what can K(lambda) tell you ?
-K has no recombination so it can tell you if the phages are on the same gene by complementation
-There on the same gene when lysis doesn’t happen (no complementation)
-Different genes when no lysis (complementation) different genes
what can B strain tell you?
-B strain tells if they are the same mutation or different since recombination can happen
-Plaques form =different mutations making recombination possible
-No plaques= same mutation making it difficult for recombination
When infecting B strain is this a genetic screen or selection?
Screen because both are infected and successfully liced
When infecting K strain is this genetic screen or selection?
Selection because only WT could be successfully liced
