1. preheat HCl for 15 mins at 60 degrees before adding root tips for a further 5 minutes.
2. remove roots from the acid and rinse well with distilled water. Proceed to cut off root tips and place in a vial containing the stain.
3. place in water bath for 5 mins before removing with forceps and placing one onto a microscope slide.
4. tease the root tip to make it as thin as possible and then add cover slip. Observe under microscope.

HCl is an irritant to eyes and skin, Acetic orcein stain is corrosive and causes burns, glassware and scissors.
