bunsen flame, loop in hot til red for a second and cool for 20 seconds

lid face down, rub culture loop on small section and sterilise and cool loop. rotate plate and do cross stitch and wiggle in the middle to drag the colonies

what is alpha and beta haemlysis
alpha: partial: each colony is surrounded by a zone of incomplete haemolysis: green pigment and reduced product of haemoglobin e.g. strep pneumonia
beta: completes zone clear in which blood is completely lysed e.g. strep pyogenes
