HISTOPATH LAB (ADH- MNT-STN) Flashcards
(35 cards)
is a substance which can be smeared onto the slide so that the sections
stick well to the slides
ADHESIVE
Slides must always be __________ and _______
and stored and handled correctly
grease, dust-free
If slides are not stored properly, _______ and ________ grows which causes false-positive results after gram
staining
bacteria and fungi
Most commonly used adhesive
ALBUMIN
Albumin is prepared through
- mixing equal parts of glycerin, distilled water, and white of eggs
- then filtered through a coarse filter
- and a crystal of Thymol is added.
One disadvantage of using albumin
it retains some of the stains and gives a dirty
background
Meyer’s egg albumin formula
- Egg white 50cc
- Glycerin 50cc
- Filter and add about 100 mg Crystals of thymol
Added to adhesive to prevent the growth of molds
Thymol
Disadvantage of Meyer’s Egg Albumin
Too thick adhesive may take up stains causing a color blue background.
How to remedy the disadvantage of Meyer’s Egg Albumin
Prepare slides again.
usually a syrupy fluid
applied between the section and the coverslip
after staining, setting the section firmly
preventing the movement of the coverslip
mounting medium
it bonds specimen, slides,
and the coverslip together with a clear durable film
mounting medium
Mounting prevents _______ and _________
Oxidation, scratches
refractive index of tissue
1.515
It is a transparent, almost colorless oleoresin
that adheres firmly to glass and sets to a hard
consistency without granulation
Canada Balsam
Canada Balsam RI value
1.524
Canada Balsam recommended for whole
mounts and for thick sections because it _______________________
does not shrink much
Disadvantage of Canada Balsam
Expensive
Canada Balsam is sourced from ________________
Canadian Tree, Abus Balsamea
CHARACTERISTICS OF A
GOOD MOUNTING MEDIUM
- be colorless and transparent.
- be freely miscible with xylene and toluene.
- not dry to a non-stick consistency and harden
relatively quickly. - protect the section from physical damage and
chemical activity (oxidation and changes in pH). - be resistant to contamination (particularly
microorganism growth). - not cause shrinkage and distortion of tissues.
- not leach out any stain or affect staining
- not change in color or pH.
- be compatible with the adhesive in use.
- set without crystallizing, cracking, or shrinking
(or otherwise deform the tissue being mounted)
and not react with leach or induce fading in
stains and reaction products (including those
from enzyme histochemical, hybridization—).
The process of sealing the margins of the
coverslip to:
★ prevent the escape of fluid or semi-fluid
mounts and evaporation of mountant
★ fix the coverslip in place
★ prevent sticking of the slides upon
storage.
Ringing
easier to use and less bubble formation
EPON: epoxy resin
The process is whereby the tissue components
are made visible with a dye or staining solution
STAINING
main principle of Staining
Done to visualize cells
