  • serum from animal is placed on a well
  • +ve control is placed on anther
  • soluble ag goes on top 3rd
  • formation of white line precipitin bw sample well, if there is a well (Hole) this is a positive test

Immunochromatography -pen side test
used to detect ag or ab
for ag detection
sample containing Ag flows through porous strip containing dried labeled ab and forms immune complexes, ab is labelled with gold, fluid with immune complexes flow through the detection zone and binds immobilising abs
for positive results there will be a colour change

For Immunohistochemistry what does it detect and explain the steps

A

Immunohistochemistry
detects Ag in situ in tissues
shows presentce of T cells in lamina propria and epithelium of canine duodenum
tissue fixed and thin sections are placed on the microscope slides
cells binding the ab recognise canine CD3 are exposed to peroxidase- labelled specific anti globulin
presence of peroxidase will turn brown

A

Flow cytometry
can tell you about the size and the granularity and internal complexity
-fluorescent labels cell surface or intraceullular ag using fluorescent ab
multiple ab can be used to stain multiple markers on a single cell
can determine relative % of immune cells
