Macrophage / Mononuclear Cell Flashcards
(16 cards)
Charactestic
- Dominant cell in chronic inflammation
- driver from circulatory blood , in tissue = macrophage
Names
- Bone: osteoclasta
- Liver: kupffer cell
- CNS: microglial cell
- Lymph node, spleen: sinus histocyte
- Lung: alveolar macrophage, in ruminant: pulmonary intravascular macrophage
Comprise
- mononuclear phagocyte system
- old name: reticulo endothelium system
Function, act as:
- filtration particular matter, microbe, senescent cell.
- act a sentinels to alert specific components of adaptive immune ( T,B lymphocytes ) to injurious stimuli.
Half life of circulatory monocyte about ________, under influence ________ and _________ they migrate to injury site within _________ after _____________
1 day
Adhesion molecule, chemotactic factor
24-48 hours
Onset of acute inflammation
Monocyte reach extravascular tissue and transformation to __________ which has __________ and greater __________ than monocyte
Large macrophage
Longer half life
Capacity for phagocytosis
Activated macrophage, result in:
- Increase cell size
- Increase metabolism
- Greater ability to kill ingested microbe
- Increase content of lysosomal enzyme
Light microscope, macrophage appear:
- large
- abundant and clear cytoplasm, pink
- single oval to polygonal, reniform nuclease, eccentric
Activated macrophage can differentiate to
Epitheliod macrophage —-> multinucleated giant cell
Activation signal for activated macrophage:
- Bacterial endotoxin and product of microbial.
- Cytokines produced by T lymphocyte ( IFN-γ ).
- ECM proteins.
- Mediator produced during acute inflammation.
After activation, macrophage:
Secrete active product resulting in tissue injury and fibrosis
Product: AA,NO,ROS metabolites , cytokines, growth factor, acid and natural proteases, plasminogen activator
Epitheliod cell :
- shape
- large than activated macrophage.
- abundant cytoplasm
- cell membrane polygonal to elongated
- forming sheets resemble squamous epithelial
Epitheliod cell :
- function
- diminished phagocytosis capacity but contain large amount of Golgi, RER, vesicle and vacuoles.
- has extracellular secretion.
Multinucleated giant cell
- syncytial cell formed by fusion two or more activated macrophage into large cell have two of more nuclease
Nuclei can be distributed in cell in haphazard manner or aggregated in the center of cytoplasm. This called__________
Foreign body multinucleated giant cell
Nulcei may arranged in horseshoelike semicircle at periphery this is called________
Langhans type giant cell
