Q

What is the role of MgCl2 in PCR

A

Mg bind single nucleotide and acts as a cofactors for DNA polymerase.
Too much MG stabilizes dsDNA and prevents desaturation of strands.

Q

What is the role of ethidium bromide in gel electrophoresis

A

Large flat basic molecule that can insert in DNA strand. It forms flourescent complexes with the nucleic acids and can be visualized under UV light

Q

What is the loading buffer made up of

A

Sucrose - makes sample dense and allows it to sink into well

Bromophenol blue - gives color for visualisation. Migrates at roughly 50bp
