Signs of liver toxicity?
Jaundice, abdo pain, vomiting/nausea, dark urine
Side effects of methotrexate
Pulmonary toxicity

What is used to reduce mucosal and GI side effects of methotrexate?
Folic acid (To be taken on the days methotrexate isn’t taken on)
Action taken if diarrhoea or stomatisis occurs
Withdraw treatment as can be first signs of GI toxicity
What is stomatitis ?
Inflammation/sorness of mouth
Contraception advice for men and women taking methotrexate?
Effective contraception during and 6 months after treatment
Breastfeeding on methotrexate?
No
Reversal agent for methotrexate
Folinic acid
Monitoring requirements for Methotrexate
LFTs, U&Es, and FBC every 1-2 weeks until stable, then every 2-3 months.
CI of methotrexate (4)?
Ascites (accumulates in ascites), Immunodeficiency conditions, pleural effusion, active infection
Interactions with Methotrexate
Trimethoprim/ Co-trimoxazole (folate antagonists), NSAIDs, PPIs, CYP450 inhibitors)
Increased risk of blood disorders with which medications?
Phenytoin, Clozapine, Trimethoprim/ Co-trimoxazole (folate antagonists)
