Complex & Minimal defined
What is Complex or Rich media?
Contains non-defined components
- LB medium has:
– Bactotyptone (protein), Bacto yeast extract, NaCl (osmolarity balance), pH of 7
What is Minimal defined media?
Contains known components
- M9 medium has:
– Glucose, NaCl, MgSO4, CaCl2….. , pH of 7
Most common way to determine/measure viable cells?
Staining & measuring colony forming units (cfu)
What is a batch culture?
A set amount of time that a bacteria is grown to obtain results
Define Lag phase
No growth - in prep for growing
Define Log phase
Exponential phase - growth approximates an exponential curve
Define Stationary phase
Cells stop growing but are not dying just surviving
Define Death phase
Cells die
Can bacteria be grown in batch culture of liquid?
Yes
What is doubling time?
Calculated from exponential growth in the log phase (denoted by ‘g’)
What is continuous culture?
Constantly adding fresh and drawing off old media to maintain exponential growth

Thermophiles (& Extreme thermophiles)
What organisms thrive in (relatively) normal temperatures?
Mesophiles
