what is microscopy?
the preparation and examination of microscope slides
what is cell fractionation?
the process of seperating the key components and organelles of cells
what needs to be controlled before homogenisation can be carried out?
it must be an ice cold, isotonic, buffered solution
what is homogenate?
the blended cell solution
what is filtration?
the process of filtering the blended cells to remove the broken cell membranes
what is ultracentrafugation?
the process of spinning the filtered cells in a centrafuge at increasing speeds to seperate out each organelle
what is the supernatent?
the liquid left behind once a cell solution has been supercentrifuged and the pellet removed
what is an artefact?
an object in a photomicrograph that shouldn’t be there
what is a photomicrograph?
the image a microscope creates
what is a vacuum?
an area found inside electron microscopes with no molecules, where sound cannot travel
what are some examples of microscope artefacts?
streaking/halos
what is resolving power?
resolution, the minimum distintance apart two points have to be so you can distinguish them
what is magnification?
the number of times bigger a microscope makes an image
what are the two types of electron microscope?
transmission (TEM) and scanning (SEM)
how does a TEM create an image?
it detects where electrons pass through the dead cell to create an image based on density
how does an SEM create an image?
it records the angles electrons bounce off the 3D shape to create an image
