What does the brca mutation do?
Increase breast cancer risk
What does the HER2-positive mutation do?
poor response to chemo
What are restriction endonucleases?
Endonucleases are isolated from bacteria to be used by scientiscts
Dna is cleaved into smaller fragments by restriction enzymes
They cleave from 4 to 8 nucleotides
these are called restriction sites
What are palindromes?
Top and bottom strands have the same exact sequence when read from 5’ to 3’
What is an application of DNA cloning?
Human insulin production
How does DNA cloning take place?
A plasmid that has antibiotic resistance is cleaved by restriction endonucleases
the DNA of intrest is also cleaved
DNA ligase is used to combine the cleaved plasmid with the cleaved DNA
The recombinant DNA (Hybrid plasmid) is insterted into the bacteria
The bacteria is then placed in a culture with antibiotics to purge any bacteria that doesn’t contain the hybrid plasmid
After the bacteria containing the hybrid plasmid multiplies, it is placed in a tube an killed to extract the multiplied DNA.
The hybrid plasmids are then cleaved again to release the cloned DNA.
