What are probes and what is its function?
A short piece of complimentary DNA or RNA
Recoginzes a DNA sequence and binds to it
it changes color/emits light when it binds (Molecular beacon)
Why aren’t probes used anymore?
Bc they break extremly easily to the presnce of RNAses
How does the three step process of probes take place?
dioxygenin or biotin are added, they bind to the dna of interest
Anti-dioxygenin or avidin is added which then binds to dioxygenin/biotin
Lastly, an enzyme is added, antidioxygenin: Horseradish peroxidase or alkaline phosphate
Biotin –> avidin
What is the defention of the FISH probes test?
The test uses probes to detect the amplification or deletion of certain genes
Flurosence in situ hybridiazation
What is the FISH test usually used to detect and give me an example?
Usually Used to detect STDs
Example (Not releated to STDs):
HER2 gene for breast cancer
Bc there are multiple red dots, it is amplified
What is dot blot test and how is it done?
it is a test that uses probes to detect a mutation to know if a patient is a carrier, normal, or infected
There are 2 papers, 1 for normal alele and the other for mutated alele
The papers tell you whether or not there is a mutated/normal alelle based on which one reacted
