Chromosomal region where replication initiates.
DnaA
AAA ATPase protein that oligomerizes at the bacterial origin and unwinds DNA to initiate replication.
AAA ATPase
Family of proteins that can bind either ATP or ADP, act as ATPases; usually more active in the ATP-bound form.
Consensus Sequence
the ideal DNA sequence that a protein interacts with.
DNA Unwinding Element (DUE)
A specific sequence in oriC that is unwound by DnaA.
Temperature Sensitive
A type of mutation that displays a wild-type phenotype at a low temperature (e.g. 30C) but a mutant phenotype at a higher temperature (e.g. 42C).
FIS (Factor for Inversion Stabilization)
DNA binding protein that bends DNA; blocks DnaA and IHF in oriC.
IHF (Integration Host Factor)
DNA binding protein that bends DNA
SeqA
DNA binding protein that bind hemimethylated GATC sequences.
RIDA (Regulatory Inactivation of DnaA)
Activity of replisome to stimulate DnaA ATPase activity, thus converting DnaA-ATP to DnaA-ADP. Conferred by Hda protein.
DnaA titration
A mechanism for sequestering DnaA-ATP, by having it bind to non-oriC sequences throughout the chromosome.
DARS
DNA sequences that bind DnaA and stimulate it to release ADP; apo-DNA then acquires an ATP.
