PCR steps
A
- Samples are heated to 94-96˚C for 30s or more to denature the target DNA, separating it into single strands
- Temperature is lowered to 50-65˚C for 30s to allow left and right primers to anneal (make double-stranded) to their complementary sequences. The primers are designed to bracket the DNA region to be amplified
- The temp. is raised to 72˚C for 30s or more to allow Taq polymerase to attach at each priming site and extent (synthesise) a new DNA strand
