CP5 method
A
- prepare boiling tube of 1M HCl and place in 60˚C water bath for 10 mins
- remove tips of the garlic/onion roots (about 1cm) and place in warmed HCl for 5 mins
- rinse tips in cold water using a pipette and blot dry with a paper towel
- cut approx. 2mm off the tip and place onto a microscope slide
- add a drop of toluidine blue
- squash stained root tip on a glass slide using a blunt instrument
- view slide under the microscope
- draw cells undergoing mitosis
