CP5 method

A
  • prepare boiling tube of 1M HCl and place in 60˚C water bath for 10 mins
  • remove tips of the garlic/onion roots (about 1cm) and place in warmed HCl for 5 mins
  • rinse tips in cold water using a pipette and blot dry with a paper towel
  • cut approx. 2mm off the tip and place onto a microscope slide
  • add a drop of toluidine blue
  • squash stained root tip on a glass slide using a blunt instrument
  • view slide under the microscope
  • draw cells undergoing mitosis
