Methotrexate
Class: DMARD (conventional)
Indication: RA + cancer!
MOA: Prevents AICAR conversion to FAICAR, allows more adenosine to be used, that decreases inflammation and pain
Adverse Effects: highly toxic
Adalimumab
Class: DMARD (biological)
Indication: RA
MOA: Blocks TNFa from binding to cells, decreases IL levels (1, 6, 8).
Adverse Effects: immunosupression
Allopurinol
Class: Xanthine Oxidase Inhibitor
Indication: Gout
MOA: blocks xanthine oxidase, which reduces the amount of purines being converted to uric acid.
Adverse Effects:
Risedronate
Class: Bisphosphate
Indication: Osteoporosis
MOA: deposits in bone by binding to calcium, gets uptaken with osteoclasts when absorb bone, causes apoptosis, decreases bone resorption.
Adverse Effects: Esophagitis, osteonecrosis of the mandible.
Denosumab
Class: RANK-L Inhibitor
Indication: Osteoporosis
MOA: binds to RANK-L which allows the maturation of osteoclasts = reduced numbers.
Adverse Effects: injection site redness
Romosozumab
Class: Sclerostin inhibitor
Indication: osteoporosis
MOA: binds to sclerostin, which stops it from binding to osteoblasts and inhibiting their wnt signalling pathway, which is essential for their activity.
adverse effects: injection site redness
