Microscopes👩🏾🔬 Flashcards
What is the equation of magnification?
Size of image/real object size
What are the rules for a good scientific drawing?
-big enough not small
-dotted lines to show it is 3D
-labelled diagram
-titles
-state magnification in drawing
-no shades
What is the magnification?
Is the number of times a cell/object diminishes or enlarged
What is resolution?
Is the distance between 2 objects to be seen as separate for a clear visual.
How an optical light microscope work?
Light source(lamp) goes through 3 different lenses
-condenser lens
-specimen
-objective lens
-eyepiece lens
How to calibrate an eyepiece graticule?
Setup: Line up the eyepiece graticule scale to the stage micrometer scale
-record how many divisions of eyepiece graticule correspond to a know length of stage micrometer
-know length about stage micrometer/number of increments(lines) on eyepiece graticule.
Remember to convert all unit to micrometer meter
Does the eyepiece or stage micrometer move?
-Move:stage micrometer,10micro.m long
Divides in 100 sub-division
-Same/constant:eyepiece graticule
-10 micro.m long
-1micro.m subdivisions
-Smallest subdivision of 0.1 micro.m
What is the advantage + disadvantages of light microscope?
-portable
-easy to use
-can see dead or alive specimen
-cheap
Disadvantages
-lower resolution and magnification
What is the max resolution and magnification and what can’t it (light microscope)not view and how it forms images and focuses?
-Max resolution:0.2 micrometer
-Max magnification:*1500
-Can’t view: smaller organelles like mitochondria, ribosomes,ER,lysosomes.
Scale from cm to nm?
cm * 10= mm
mm * 1000= micrometer
Micrometer * 1000 = nm
What does light and T.E.M and S.E.M offer?
Light:gives a magnificated image and lower resolution and magnification
TEM:transmission electro microscope, sees inside, higher resolution and it is black and white
SEM:scanning electron microscope,3D view (structure is given but not inside)and higher resolution and magnification.
How does the optical light microscope form an image and how it makes it to focus?
Uses light to form image and focus uses glass lenses
How do we identify starch grains in plants cells?
Is the Iodine in
Potassium iodine solution to identify them
