Conventional DSBR - Alternative Non-homologous End Joining Flashcards
(6 cards)
1
Q
What is the purpose of an alternative NHEJ pathway?
A
It acts as a fail-safe mechanism when the cell is defective for classic NHEJ component Ku.
2
Q
What is the disadvantages with using this pathway?
A
Alternative NHEJ takes longer than classical NHEJ and it is highly error prone because it can cause inversions and chromosomal rearrangements along with deletions.
3
Q
Step 1 - A-NHEJ
A
Poly(ADP-Ribose)Polymerase 1 (PARP1) binds break points.
4
Q
Step 2 - A-NHEJ
A
PARP1 recruites Mre11-Rad50-Nbs1 complex.
5
Q
Step 3 - A-NHEJ
A
MRN complex and CtIP resect the breaks.
6
Q
Step 4 - A-NHEJ
A
DNA Ligases I and III join DNA ends together.
