How do defects in DNA repair mechanisms lead to cancer development and what are some examples?
A
Genetic mutations and chromosome abberations accumulate. Some examples are Ataxia-Telangiectasia, Fanconi Aneamia and Li-Fraumeni Syndrome.

What are the goal of cancer therapy?
A
To cause DNA damage in cancer cells and inhibit DNA proteins leading to genomic instability that will cause apoptosis.

How are SSB repairs and the BER exploited in cancer treatment?
A
PARP inhibitors compete with nicotinamide for the catalytic site of PARP molecules and without nicotinamide PARylation cannot occur and PARP1 cannot function to repair DNA.
