What are the 2 major sites of glycogen storage ?
- Liver- maintains blood glucose levels
- Muscle- provides local energy for muscle contraction
What proteins facilitate glucose uptake from the blood ?
- GLUT 2- present in liver and B cells of pancreas. Low affinity, high capacity
- GLUT 4- present in muscle and fat cells. It is insulin dependent
What is glycogenesis ?
The process of synthesising glycogen- the storage form of glucose
What are the steps of glycogenesis ?
- Glucose is converted into Glucose 6-phosphate by hexokinase in muscle or glucokinase in the liver
- Glucose 6-phosphate is converted into Glucose 1-phosphate by Phosphoglucomutase
- Glucose 1-phosphate is activated by enzyme UDP-glucose pyrophosphorylase, which produces UDP-glucose
- Glycogenin auto catalyses the addition of glucose to itself to start the glycogen chain.
- UDP-glucose is added to glycogen chain to form alpha 1,4 glycosidic bonds using enzyme glycogen synthase
What is the key regulatory enzyme of glycogen synthesis?
Glycogen synthase
Active form: desphosphorylated (stimulated by insulin)
Inactive form: phosphorylated
What forms alpha 1,6 glycosidic bonds in glycogenesis ?
A branching enzyme
Describe the steps of glycogenolysis
- Glycogen phosphorylase cleaves alpha 1,4 glycosidic bonds by adding inorganic phosphate to produce Glucose-1-phosphate
- A debranching enzyme removes the 1,6 glycosidic bonds
- Glucose 1-phosphate is converted into Glucose 6-phosphate using enzyme Phosphoglucomutase
- In the liver Glucose 6-phosphatase breaks Glucose 6-phosphate into glucose which can enter the blood
What is the key regulatory enzyme in glucose breakdown ?
Glycogen phosphorylase
How are glucose synthesis and breakdown regulated ?
By hormone triggered (glucagon or adrenaline) cAMP cascade acting through Protein kinase A
What are examples of glycogen storage diseases ?
- Von Gierke’s disease
- Cori’s disease
- McArlde’s disease
What is Von Gierke’s disease ?
Type 1.
Glucose-6-phosphatase is missing
Hypoglycaemia occurs
What is Cori’s disease ?
Type 3
Deficiency of Glycogen debranching enzyme.
What is McArlde’s disease ?
Type 5
Muscle phosphorylase missing
Muscle cramps occur
What does protein phosphatase 1 do ?
Reverses the effect of protein kinase A
