What is microscopy?
The use or investigation of a microscope to study tiny objects.
Who first made eyeglasses?
Salvino D’Armate from Italy in the 13th century.
What are ‘flea glasses’?
Early magnifiers with 1 convex lens used to inspect insects.
Who invented the first compound microscope?
Zacharias and Hans Jansen in the 1590s.
Who was the first to use a real microscope?
Anton van Leeuwenhoek in the late 1600s.
How did Van Leeuwenhoek make powerful lenses?
He polished over 500 lenses for greater magnification.
What does a light microscope use?
Light and glass lenses to magnify an image.
What does an electron microscope use?
Electrons for much higher magnification.
What is the function of the eyepiece lens?
Lens closest to your eye
What is the function of objective lenses?
To magnify the image further with 3+ strengths.
What is the function of the diaphragm?
Controls the amount of light that hits the slide.
What is the function of the coarse focus knob?
Used to bring the object roughly into view.
What is the function of the fine focus knob?
Used to sharpen the image clearly.
How do you calculate total magnification?
Eyepiece lens × Objective lens.
What’s the total magnification for 10x and 40x?
400x.
Why draw microscope slides in pencil?
Because shading and coloring are not needed; it should be clear and simple.

Magnification
What is the first step in preparing a microscope slide?
Take a sample of the tissue or specimen you wish to observe.
What do you place the specimen on?
A clean glass slide.
Why do we add a dye or stain to the slide?
To make the specimen or its structures easier to see under the microscope.
What is the coverslip used for?
To flatten the specimen and avoid air bubbles.
Why should you dab the slide with tissue?
To remove any excess liquid.

Observe it under the microscope.
