What are the characteristics of Pasteurella multocida?
Gram neg coccoid rods with bipolar staining
May be capsulated
Facultative, 35°C
BAP- grey, translucent, gamma
MAC- no growth
Why does Pasteurella multocida not grow on MAC?
Inhibited by bike salts
What are the ID tests for Pasteurella multocida?
Ox pos (with tetramethyl)
Glc fermentation
Indole
Ornithine, sucrose, urease
Susceptible to low concentrations of penicillin
What is the significance of Pasteurella multocida?
NF in the respiratory tracts of dogs and cats
Hemorrhagic septicaemia in cattle
Avian cholera
Infection from animal contact
Septicaemia leading to osteomyelitis, meningitis, other infections
Respiratory tract infections in those with preexisting pulmonary disease
What can be used to treat Pasteurella multocida?
Penicillin
Tetracycline
Some cephalosporins
Resistant to- aminoglycosides
What are the characteristics of Bordetella?
Very small gram neg coccobacilli
Older- pleomorphic
Pale pink staining
Obligate aerobes
What are the species of Bordetella?
pertussis, parapertussis, bronchiseptica
How can Bordetella staining be improved?
Longer counter stain
Dilute carbol fuchsin
How are specimens for whooping cough diagnosis collected?
Pernasal nasopharyngeal specimen
Nasopharyngeal secretions
What transport medium is used for Bordetella specimens if they are plated immediately?
Regan and Lowe (RL)- charcoal
Casamimo acids- direct smear prep for Ab staining
What planting media is used for Bordetella and what are their characteristics?
Bordet Gengou (BG)- potato infusion, sheep blood, antibiotics, short shelf life
RL- charcoal agar, horse blood, cephalexin, better shelf life
How long are Bordetella specimens incubated for?
5-7 days
See colonies at 5-7
What are the characteristics of Bordetella colonies?
BG- small, pearly grey, drops of Hg
RL- small, done shaped, white-grey, glistening
What is the ID for Bordetella pertussis?
Slow growing
Isolated on B. pertussis media
No growth on BAP/MAC/CHOC or regular culture media
Assaccharolytic
What is the rapid diagnosis for Bordetella pertussis?
DFA stain
Direct smear treated with fluorescent labelled pertussis antiserum
What is the significance of Bordetella pertussis?
Infection from droplet inhalation
Incubation- 5-7 days
Catarrhal stage- mild UPT infection
Paroxysmal stage- caused by toxins, airway obstruction, aspiration, pneumonia, neurobiological complications, marked lymphocytosis
Convalescent stage- several weeks
No passive protection from maternal Abs
