Thick peptidoglycan layer- retains crystal violet stain
(Purple under microscope )

Thin peptidoglycan layer- alcohol will wash crystal violet out

Saffron stains cells red

Have layers of lipopolysaccharides - protect cell from lysosomes and antibodies
