what is the giemsa stain
-made by gustav giemsa a chemist and bacteriologist
-mix of an acidic (eosin) and two basic dyes (methylene blue/azure B)
-specidic for phosphate groups on dna (stains anything with dna)
giemsa stain is used for staining for
-blood smear (leukocyte cells) and parasites like plasmodium, trypanosomes, microfilariae and leish
how is the blood collected for a giemsa stain
-finger, ear lobe or toe for kids
what is the difference between thick and thin blood films for malaria diagnosis
-thick: parasite detection for low parasitemia
-thin film: used for confirmation and species identification
thick blood film prep
-touch the blood drop with a clear slide or 2 drops of blood (30-40 ul)
-using the corner of another slide, spread the blood into the shape of a circle of 1-1.5 cm2
thin blood slide prep
-touch the blood drop with a clear slide
-hold the slide and with another slide at 30-45 degree angle touch the drop and wait until the blood spread along the edge of the second slide
-rapidly and smoothly push slide forward
difference between p vivax and p falciparum multiple infection in rbc
vivax: only has one ring
valciparum: common aka there are more than 1 ring
difference between p vivax and p falciparum parasitemia
vivax: low
falciparum: high
difference between p vivax and p falciparum ring stage
vivax: relatively large usually 1 chromatin dot
falciparum: may have 1 or 2 chromatin dots
difference between p vivax and p falciparum gametocytes stage
vivax: round
falciparum: sausage shaped
what do monocytes look like
kidney shaped nucleus
what do neutrophils look like
multiple lobed 3-5 nucleus
what do lymphocytes look like
one round nucleus, little cytoplasm
what do eosinophils look like
2 lobed nucleus; red granules in cytoplasm
what do basophils look like:
large blue granules in cytoplasm
