Q
What are the four possible mutations in Lac- bacteria?
A
1) Mutation in lacZ
2) Mutation in promoter (deletion)
3) Mutation in LacI
4) Lac Y mutation
2
Q
Mutation in Lac Z
A
lac Z gene encodes B-Galactidase so none would be produced
3
Q
Mutation in Promoter
A
No genes for the operon would be produced
4
Q
Mutation in LacI
A
Lac I makes repressor so no repressor meaning no transcription
5
Q
What is ONPG used for when doing the B-Galactidase assay?
A
ONPG acts as the substrate for B-Galactidase (usually this would be lactose but we used IPTG instead which is not a substrate) ONPG results in yellow coloring for assay when added to induced lac operon
