Bacteriphage as potential therapy
When viruses were discovered this was super popular but quickly died out
Penecillin
replaces viruses as the primary therapy against infections
Cryo-EM
allows detailed 3D reconstruction of phage particles (phage T7)

What does a plaque contain on a lawn?
contains progeny of single infecting virus (a single lineage)

can go through lytic cycle or lysogenic cycle
What color plaque do lytic produce versus temperate phage?
Lytic-clear
Temperate- turbid plaques
What is a maltose porin (LamB)?
a porin created in cell membrane of E.coli when grown in maltose
what decides wheather the lamba will enter the lytic or lysogenic cyce?
cll and cro
When cll predominates what happens?
Lysogen formed
When cro predominates:
Lambda enters lytic cycle
What does stress do?
induces lytic cycle through SOS response (actiates RecA which inactivates LexA repressor)
How does lamba inssert into chromosome?
Site specific recombination
