Module 2 Flashcards

1
Q

Describe basic microscopy

A

Thickness of section: too thick can lose ability to discriminate structures
lighting: can generate more contrast by altering amount and angle of light

Q

What is magnification and resolution?

A

Resolution is the distance two objects need to be apart in order for us to see in between them or the width of an object that it needs to be in order to make it visible
-a lense that is closer will have a larger light angle and thus better resolution
-Shorter wavelength = smaller R which equals higher resolution
-Oil immersion has higher refractive index than air providing better resolution

Q

what is absorbance vs transmittance

A

Transmittance: the proportion of light that the sample allows to pass (usually %)
Absorbance: the amount of light the sample blocks, calculated as A = Log (1/T) OR -Log(T) values of 0-2 are typical
-if given abs to solve for T, take 10^-A
