Q

procedure for reticulocyte count

A

Place three drops of reticulocytes stain in a small test tube.
Add three drops of well mixed whole blood to the tube containing the stain.
Mix the tube and allow to stand at room temperature or incubate at 37C for 15 minutes
After 15 minutes, mix the contents of the tube well and prepare smears and allow it to dry.
Examine the smear under the oil immersion objective.
Count the total number of reticulocytes found per 1,000 RBCs

16
Q

why edta is the ideal tube top?

A

it is stable for the span of 24 hrs, however, the retics will decrease within 6-8 hrs after collection

17
Q

3 components that are sometimes being misidentified as retics

A

pappenheimer
heinz bodies
howell jolly

18
Q

why papperheimer is being misidentified as a retic?

A

because of the presence of iron that appears like rna after staining

19
Q

why heinz bodies are being misidentified as a retic/

A

it composed of denatured rrbc

20
Q

why howell jolly is being identified as a retic

A

it has the remnants of rna

21
Q

what are the 2 interferences of retics count?

A

heparin and the high blood glucose level

22
Q

why high blood glucose level is included in the interferences of retics count?

A

will cause pale staining to the supravital stain

23
Q

conditions where retics are found elevated or high.

A
  • hemolytic anemia
  • IDA
  • thalassemia
  • sideroblastic anemia
24
Q

conditions where retics are found low or decrease

A
  • aplastic anemia
  • BM failure
  • kidney failure
