What is UV-visible spectroscopy?
A technique for analysing the absorption of UV or visible light by molecules, causing excitation of electrons
What order does UV-vis absorption excite electrons in ?
From HOMO to LOMO
Highest occupied molecular orbital
What do * and n denote ?
- is anti-binding
n is non binding
What is emission ?
When an electron transitions from a higher energy state to a lower energy state
What are the applications of UV-vis spec ?
- Quantitative measurements of coloured and fluorescent substances
- Tagging of targets. Eg. DNA sequencing
- Histo chemistry and imaging
What are chromophores ?
Structural units absorbing light
What are auxochromomes ?
Modify intensity or wavelength of absorption
What is a hypsochromic shift ?
To shorter wavelengths (blue shift)
What is bathochromic shift ?
To longer wavelength (red shift)
What is hypchromic shift ?
Decreased intensity
What is hyperchromic shift ?
Increased intensity
