Investigating enzymatic reactions Flashcards

1
Q

how do you investigate the effect of pH on enzyme activity

A

1 - drop of iodine solution into every well of a spotting tile
2 - place a bunsen burner on a heat proof mat and then a tripod and gauze over the burner.
3 - put a beaker of water on top and heat it until 35 C and keep constant
4 - add 1 cm ^3 of amylase solution and 1 cm^3 of buffer solution with a ph 5 with a syringe into a boiling tube then into the water and wait 5 minutes
5 - add 5 cm^3 of starch solution with a clean syringe them mix it and start the clock
6 - record how long it takes for the amylase to break down ising continuous samples
7 - when the iodine solution remains browny-orange there is no more starch
