What is core practical 11? (1)
Investigate the presence of different chloroplast pigments using chromatography
What is the first stage of this practical? (1)
Cut spinach leaves into small pieces with scissors and place into a mortar with a sprinkling of sand.
What is the second stage of this practical? (1)
Use a pestle to grind the mixture, add a small amount of propanone to the paste as you grind.
What is the third stage of this practical? (1)
Allow the mixture to settle and draw up the dark green chlorophyll solution with a small pipette, place this in a small, sealed tube and allow any solids to settle. The mixture can be filtered but you need to add more propane as some is absorbed by the filter paper.
What is the fourth stage of this practical? (1)
To prepare a chromatogram, cut a piece of chromatography paper to fit the container, use a pencil to draw a line 1cm above the bottom
