What are the steps of a acid fast stain?

A

Put bacteria on slide, air dry and heat fix, put a paper towel on top of sample and steam the smear for 5 minutes, continuously adding carbol fuchsin. remove the paper towel and rinse with water, and then rinse with acid alcohol for 15 seconds, counterstain with methylene blue and look under microscope

A

Put sample on slide (air dry and heat fix), put paper towel on slide, steam slide for 5 minutes while putting Malachi green on it, remove paper towel and rinse with water. Use safranin for 1 minute and wash and dry

A

touch tape to mold and tape it to the slide, on drop of cotton blue on top of tap and put cover slide on top of blue. Put over flame until blue starts boiling and look under the microscope.

A
