(27 cards)
what are the side effects associated with the administration of monoclonal antibodies ?
infusion reactions , antigen antibody binding associated with cytokine release
what is the MOA of bevacizumab ?
inhibits the formation of new blood vessels through inhibition of VEGF
what are the uses of bevicazumab ?
RCC
CRC
NSCLC
angioproliferative retinopathy
what are the side effects associated with bevacizumab ?
cardiovascular side effects
hemorrhage and blood clotting
impaired wound healing
what are the EGF inhibitors ?
cetuximab and panitumumab
they bind to EGFR - which is a tyrosine kinase receptor
what are the side effects associated with EGF moonoclonal antibodies ?
associated with acne , elevated LFT and diarrhea
what are the clinical uses of cetuximab ?
metastatic colon cancer ( wild type RAS)
head and neck cancers
if a colon cancer patient is being treated with cetuximab with no apparent improve , what is the most likely cause ?
usually because its a K-ras mutation
only affective in the wild type of K RAS
what are the PD1 inhibitors ?
pembrolizumab
what are the CTLA 4 inhibitors ?
impilimumab
what is the MOA of erlotinib ?
EGFR inhibitor
what is the major use of erlotinib and what is the main adverse effect ?
SCLC
AE : rash
what is the common side effect associated with all EGFR blocking drugs ?
skin rash
what is the MOA of imatinib ?
BCR ABL inhibitor
what are the main clinical uses of imatinib ?
CML
ALL
GISTS
what mutation is associated with GIST ?
c-KIT mutations
what is the CD associated with c KIT ?
CD 117
what is the mechanism of action of rituximab ?
anti CD 20 - leads to the depletion of B cells
what are the uses of rituximab ?
b cell malignancies
RA
ITP
what are the side effects associated with rituximab ?
opportunistic infections , especially associated with progressive multifocal leukoencephalopathy JC virus
why must a liver panel be done before the administration ?
hepatitis B reactivation
what is the MOA of tamoxifen ?
selective estrogen receptor modulator (SERM) , competitive antagonist of breast estrogen receptor
estrogen agonist in other tissues
when is tamoxifen used ?
used in ER positive breast cancer
what are the toxicities associated with tamoxifen ?
1- hot flashes
2- increased risk of DVT/PE , increased clotting factors
3- endometrial cancer (major risk in. postmenopausal women )
