What is a microscope slide?
A strip of clear glass or plastic onto which the specimen is mounted.
What is the first step to prepare a slide for viewing onion cells?
Add a drop of water to the middle of a clean slide.
What should be done after cutting up an onion for slide preparation?
Use tweezers to peel off some epidermal tissue from the bottom of one of the layers.
How should the epidermal tissue be placed on the slide?
Using tweezers, place the epidermal tissue into the water on the slide.
What is the purpose of adding iodine solution to the slide?
Iodine solution is a stain used to highlight objects in a cell by adding color to them.
What is the correct way to place a cover slip on the slide?
Stand the cover slip upright on the slide, next to the water droplet, then carefully tilt and lower it to cover the specimen.
What is the first step in using a light microscope to view the prepared slide?
Clip the slide you’ve prepared onto the stage.
What should be selected first when using a light microscope?
The lowest-powered objective lens.
How do you initially focus the microscope on the slide?
Use the coarse adjustment knob to move the stage up to just below the objective lens.
What should you do after moving the stage downwards to roughly focus the image?
Adjust the focus with the fine adjustment knob until you get a clear image.
What should you do if you need to see the slide with greater magnification?
Swap to a higher-powered objective lens and refocus.
What is the recommended tool for drawing observations under a microscope?
A pencil with a sharp point.
What should your drawing under the microscope capture?
It should take up at least half of the space available and be drawn with clear, unbroken lines.
What should not be included in the drawing of cells?
Any coloring or shading.
How should subcellular structures be drawn?
In proportion.
What is the formula to work out magnification?
magnification = length of drawing of cell / real length of cell.
What should be included in the drawing besides the observation?
A title of what you were observing and the magnification at which it was observed.
What is the importance of labeling features in your drawing?
To identify important features like nucleus and chloroplasts using straight, uncrossed lines.
